A comparative proteomic approach to identify defence-related proteins between resistant and susceptible rice cultivars challenged with the fungal pathogen Rhizoctonia solani A resistant rice verity Tequing and a succeptible verity Lemont was taken for the experiment. R. solani AG1-IA was used for infection. After the infection, stem of the plant was collected ans used for total protein isolation. Now extracted proteins were digested and lebled with iTRAQ. Now MS/MS and MS-TOF analysis was done and collected data was analysed using ProteinPilot v 4.2 for peptide identification. In house data with rice data from UniPort was used for analysis. Then GO and KEGG analysis was done follwed by RNA extraction and qRT-PCR analysis. In total 6560 ptoteins were identified out of which 755 were differentially expresing with more than 1.5 fold change. Proteomic and transcriptomic approaches to identify resistance and susceptibility related proteins in contrasting rice genotypes infected withfungal pathogen Rhizoctonia solani In this experiment 2 moderately tolerant and 4 succeptible rice verities were used and infected with R. solani AG1-IA (WGL-12-1). Then protein extraction, solubilization, IPG strip rehydration, Isoelectric focussing, SDS-PAGE and CBB analysis, Peptide fingerprinting and in-silico analysis were done. Differentially expressed proteins (DEPs) were from six rice genotypes identified which were found to be involved in different processes like photosynthesis, cell wall metabolism, development of cytoskeleton, pathogenic resistance etc. Prathi et al.
